MSCulture™ High Growth Basal Medium/Supplement
MSCulture™ High Growth Basal Medium/Supplement is a growth medium designed to support the efficient proliferation of mesenchymal stem cell (MSC) while maintaining high cell quality. It is suitable for MSCs derived from a variety of tissues, including bone marrow, adipose tissue, and umbilical cord matrix.
Compared with conventional MEMα and other growth media for MSC, this product provides enhanced proliferative capacity. MSCs expanded using this medium can subsequently be cultured in EV-Up™ EV Production Basal Medium/Supplement for MSC, enabling efficient, large-scale production of extracellular vesicle (EV), including exosome.

Protocol

[Note]
- For extracellular vesicle isolation, we recommend using PS affinity-based methods, such as the MagCapture™ Exosome Isolation Kit PS Ver.2 or the MassivEV™ EV Purification Column PS.
- MSCulture™ High Growth Basal Medium/Supplement should be supplemented with the serum of your choice.

Comparison of Cell Proliferation Rate
The cell proliferation rate of bone marrow-derived MSCs was evaluated using this product, and competitor’s MEMα and growth medium for MSC.


[Result]
This product demonstrated higher cell proliferation rate compared with the competitor’s MEMα and growth media for MSC.
Antifibrotic Activity of EVs derived from MSC
It has been reported that EVs derived from MSCs exhibit antifibrotic activity. Bone marrow-derived MSCs were expanded using this product or competitor's MEMα, then transferred to EV-Up™ MSC EV Production Basal Medium/Supplement for EV production. EVs were purified from the cell culture supernatant using the PS affinity method. Purified EVs (5 x 107 particles/mL) were added to human fetal lung-derived fibroblasts stimulated with TGF-β, and antifibrotic activity was assessed by measuring collagen V gene expression as a fibrosis marker.

[Result]
EVs derived from MSCs expanded with this product suppressed TGF-β-induced collagen V expression.
