Acid Phosphatase

During the quantification of vitamins B1 or B2, acid phosphatase can be used for converting the vitamins from their bound to free forms with high phosphatase activity.
This product contains various enzymes produced by the yellow koji mold (Aspergillus oryzae).

Features

  • The enzymes are the same as those in Taka-Diastase U, which is conventionally used for the quantification of vitamins B1 and B2, and can be used in the same manner.

Application

A. Preparation of enzyme solution

Dissolve approximately 1 g of this product in 100 mL purified water or buffer solution before use, adjust the pH to nearly 4.5, allow to stand in the cold for some time, and filter or centrifuge the solution to obtain the supernatant for use.

B. Reaction with sample

Extract and prepare the sample so that the content of vitamin B1 or B2 is nearly 10 μg. If the sample is a fluid extracted from dried yeast or vegetable tissue, such as soy, add 0.5 - 1 mL of 1% solution of this product (Acid Phosphatase) and incubate for approximately 5 h at 40 °C. For homogenates of animal tissues, such as pork and pig liver, add approximately 10 mL of 1% solution of this product (Acid Phosphatase) and incubate for nearly 10 h at 40 °C.

These are examples of general use. The method varies depending on the type of sample and the content of conjugated vitamin B1 or B2. Examine the conditions as needed.
